چکیده
I N THEIR SEARCH for the toxic factor in trichloroethylene-extracted soybean oil meal which can induce fatal aplastic anemia in calves,”2 cattle,3 and horses,4 McKinney et al.5 synthesized S-( dichlorovinyl)-L-cysteine (DCVC). They reported that the oral administration of this compound to calves produced leukopenia, thrombocytopenia, a relative lymphocytosis and a hemorrhagic syndrome, conditions which are also observed13 when sufficient amounts of trichloroethylene-extracted soybean oil meal are fed to calves. Although the toxic factor in this meal, which elicits this syndrome, has not been isolated and characterized, its chemical properties, insofar as they have been ascertained,57 are consistent with the view8 that it may be produced through the interaction of trichloroethylene with cysteine residues of proteins of the soybean. In conjunction with our other studies on bovine aplastic anemia, we have synthesized DCVC and confirmed its high toxicity for calves.

The role of glutathione conjugate metabolism and cysteine conjugate beta-lyase in the mechanism of S-cysteine conjugate toxicity in LLC-PK1 cells.
A cell line derived from pig kidney, LLC-PK1, was grown in a culture system in which the cells express morphological and biochemical characteristics of the proximal tubule. This model was used to investigate the mechanism of S-cysteine conjugate toxicity and the role of glutathione conjugate metabolism. LLC-PK1 cells have the degradative enzymes of the mercapturate pathway, and S-(1,2-dichlorov...

S-(1,2-Dichlorovinyl)-L-homocysteine (DCVHcy), an analogue of the nephrotoxin S-(1,2-dichlorovinyl)-L-cysteine (DCVCys), is a much more potent nephrotoxin than DCVCys both in vivo and in isolated renal proximal tubular cells. S-(1,2-Dichlorovinyl)-DL-alpha-methylhomocysteine, at equimolar doses relative to DCVHcy, is not nephrotoxic. Agents that inhibit pyridoxal phosphate-dependent enzymes (DL...
متن کاملMetabolism and toxicity of trichloroethylene and S-(1,2-dichlorovinyl)-L-cysteine in freshly isolated human proximal tubular cells.
Trichloroethylene (Tri) caused modest cytotoxicity in freshly isolated human proximal tubular (hPT) cells, as assessed by significant decreases in lactate dehydrogenase (LDH) activity after 1 h of exposure to 500 microM Tri. Oxidative metabolism of Tri by cytochrome P-450 to form chloral hydrate (CH) was only detectable in kidney microsomes from one patient out of four tested and was not detect...
متن کاملNephrotoxic and genotoxic N-acetyl-S-dichlorovinyl-L-cysteine is a urinary metabolite after occupational 1,1,2-trichloroethene exposure in humans: implications for the risk of trichloroethene exposure.
Excretion of mercapturic acids in the urine is indicative of the formation of electrophiles in the metabolism of xenobiotics. The determination of these mercapturic acids thus may be a useful method to estimate the exposure. We identified the nephrotoxic and mutagenic mercapturic acids N-acetyl-S-(1,2-dichlorovinyl)-L- cysteine and N-acetyl-S-(2,2-dichlorovinyl)-L-cysteine in the urine of worke...

Trichloroethylene (TCE) is a well-known carcinogen in rodents and concerns exist regarding its potential carcinogenicity in humans. Oxidative metabolites of TCE, such as dichloroacetic acid (DCA) and trichloroacetic acid (TCA), are thought to be hepatotoxic and carcinogenic in mice. The reactive products of glutathione conjugation, such as S-(1,2-dichlorovinyl)-L-cysteine (DCVC), and S-(1,2-dic...
